Design Notes
This part is designed to be a guide sequence for the igRNA because dsDNA of hsa_circ_0001982 (BBa_K4409008) is a very good candidate for the substrate dsDNA used in the CRISPR-Cas12 system of our project. Substrate dsDNA is used to activate the Cas12 enzyme. A large amount of dsDNA is needed in the system to achieve sensitive signals. hsa_circ_0001982 dsDNA can be obtained easily and abundantly when our detection tool for hsa_circ_0001785 and hsa_circ_0001982 biomarkers in blood and tissues is applied clinically, because blood samples tested positive by our tool are likely to be further analyzed by amplifying hsa_circ_0001785 or hsa_circ_0001982 for other tests such as gene sequencing. To form an igRNA, this part can be coupled with our trigger-RNA sensor (BBa_K4409003) which contains a loop sequence complementary to it, so that the loop sequence can block this part when no trigger is in the system.
